something is wrong??

I do the pcr yesterday, yes, there is the expect band after the gel detection although it is very weak and has some unspecific bands, but it does have. the strange thing is that when I do it again, there is nothing exclude the unspecific bands. I add all the things are same, something is wrong?

I have to check or do it again.

most people are on holidays

Now it is Merry X-MAS, most people are on holiday, I feel a litter lonely, but another thing is good that I can use all the PCR machine as I will. it is nice, I can do a lot of work now.

my experiment

I am trying to get a 5kb segment from the potato virus Y genome, I used the Phusion to amplify the fragment and used DYNAZYME (Both are from Finnzyme company) as control, there is a target band when using the Phusion, But there is nothing for DYNAZYME, so to get the long fragment the common taq enzyme usually are not good enough.

The PCR for the Long Product.

Sometimes it is difficult to got the long PCR product (such as 5kb), there are many influence facter, such as for the RT-PCR, The quality of the RNA, the Reverse Transcription, the primer, and the procedure of the PCR , and especially the Taq enzyme. but I think usually the Taq enzyme play an important role in this reaction. we need some taq which can amplify the long product, there are many kind of this enzyme.

I want to do the RFLP analysis, so i want to know how to select the enzyme? because i have a lot of sequences, i want to using it to distinguish different strain.
